To determine the requirement for a functional P-glycoprotein, we used rVVs expressing three mutated forms of the protein under the control of the T7 promoter, termed CM, NM, and DM. The introduced mutations alter one or both of highly conserved Asp residues located, respectively, within Walker B regions of either NH2-terminal (D555N) or COOH-terminal (D1200N) ATP binding/utilization sites that are believed to be involved in binding to Mg2+.
