Mutations (R449E, E458A, S460A, and V461A in B. subtilis) were introduced by oligonucleotide-directed mutagenesis of the pBL30 vector, in accordance with the method of Weissman et al.7) Using the resulting construct (pBL-SRI30) as a template, the Shine-Dalgarno sequence plus the His6-groEL SRI fusion gene were amplified by PCR using NHisSRI-F (50 -GTGACTAGTTAACAATTTCACAC-30 ; SpeI site underlined) and H30BL-R primers (see above).
