The cytosolic loop linking TMs 15 and 16 has been shown to play a role in sulphonylurea binding to SUR1, as mutation of S1237Y abolished tolbutamide block and [3 H]glibenclamide binding, whilst inhibition by meglitinide(whichresemblesthenon-sulphonylureamoiety of glibenclamide) was unaffected (Ashfield et al. 1999).
